| Background | IL-6R is a key component in IL-6 signaling, consisting of two subunits: IL-6Rα and IL-6Rβ (gp130). IL-6Rα is a type I transmembrane glycoprotein that binds IL-6 and modulates its bioactivity. IL-6Rα exists in two forms: membrane-bound (mIL-6R) and soluble (sIL-6R). sIL-6R can be released from activated CD4⁺ T cells through proteolysis, and it binds IL-6 to activate gp130, promoting cell regeneration and anti-inflammatory signaling. IL-6R plays crucial roles in immune responses, cell growth, and differentiation, as well as in acute-phase reactions and hematopoiesis. Additionally, IL-6R is implicated in the pathogenesis of various diseases, including multiple myeloma, autoimmune diseases, and prostate cancer. |
