Background | CD40 is a 48 kDa type I transmembrane protein and a member of the tumor necrosis factor receptor superfamily, expressed on B cells, dendritic cells, and monocytes. It interacts with CD40L to activate signaling pathways like NF-κB, MAPK, and JAK-STAT, regulating cell proliferation, differentiation, and immune responses. In B cells, CD40 activation supports survival, Ig class switching, and memory cell formation. In dendritic cells, it enhances antigen presentation and co-stimulatory molecule expression, promoting T cell activation. CD40 is also found in non-immune cells and some tumor cells, where its activation can induce macrophage activation and enhance antitumor effects. Thus, CD40 is a key target for immunotherapy. |
