| Background | CD19 is a B cell surface antigen belonging to the immunoglobulin superfamily, expressed on all B cell lineages except plasma cells and follicular dendritic cells (FDC). It acts as a crucial co-receptor of the B cell receptor (BCR) complex, lowering the signaling threshold for BCR activation and regulating B cell activation, proliferation, and differentiation. CD19 forms a signaling complex with molecules such as CD21 and CD81, enhancing B cell antigen recognition and antibody production. In mouse models, CD19 deficiency results in abnormal B cell development and impaired antibody generation. Additionally, CD19 is a significant target in cancer immunotherapy, especially in CAR-T cell therapies targeting B cell malignancies. |
